Merupakan anak dari pasangan Eric Lawrence 'Rick' Bernthal dan Joan Lurie.

Setelah lulus SMA, Jon Bernthal sempat berkuliah di Skidmore College, New York.

Akan tetapi keluar di tengah jalan dan memilih mengikuti pendidikan seni teater di Russia's Moscow Art Theatre.

Semasa tinggal di Moscow, Jon Bernthal bermain baseball sebagai catcher.

Selanjutnya, Jon Bernthal kembali ke Amerika Serikat untuk mengikuti pendidikan di American Repertory Theater di Cambridge dan berhasil menyelesaikannya pada tahun 2002. (1)

Karier Jon Bernthal sebagai aktor dimulai dari panggung teater.

Tercatat, Jon Bernthal telah tampil di panggung teater sekitar 30 kali sejak melakoni debut profesional pada 2002.

Di tahun yang sama, Jon Bernthal membintangi 'Law & Order: Criminal Intent' sebagai Lane Ruddock.

Debutnya tampil di layar lebar juga dilakoni di tahun yang sama dalam film 'Mary/Mary' sebagai Manny.

Pada tahun 2008, Jon Bernthal berperan sebagai Donnie Pitron dalam film 'Bar Starz'.

Titik tolak karier Jon Bernthal dimulai pada tahun 2009 saat bermain dalam film 'Night at te Museum: Battle of Smithsonian', berperan sebagai Al Capone.

Seiring berjalannya waktu, nama Jon Bernthal semakin diperhitungkan di dunia hiburan dan telah membintangi berbagai judul film.

Baru-baru ini, Jon Bernthal tampil dalam film 'The Peanut Butter Falcon' (2019) dan 'Ford vs Ferarri' (2019).
